Methodological tool for the integral analysis of histopathological changes in pancreas and kidneys of diabetic rats

Abstract

Introduction: Histopathological studies in experimental models of diabetes are usually performed using separate semiquantitative or histomorphometric methods which prevents the integral interpretation of the results.

Objective: To desing a methodological tool based on the combined application of semiquantitative and histomorphometric methods for the histopathological evaluation of changes in pancreas and kidneys of diabetic rats.

Methods: Two groups of male Sprague-Dawley rats were used: a healthy control and a diabetic group (alloxan 120mg/kg). After 15 days, pancreas and kidneys from all animals were removed and processed for histopathological examination. The semiquantitative method was applied to sections stained with Hematoxylin and Eosin (HE), while for the histomorphometric method the special Gomori's Aldehyde Fuschin (AFG) stain was used in pancreas and the HE stain in kidney.

Results: The application of semiquantitative methods showed a significant increase in the grade of insulitis (pancreas) and glomerular and interstitial damage (kidney) in diabetic animals.  By means of histomorphometric methods was demostrated a significant decrease in area of islets of Langerhans and density of beta cells (identified by the special stain AFG) in pancreas, and glomerular diameter and area kidney, in the diabetic group. 

Conclusions: The combined application of semiquantitative and histomorphometric methods allowed to integrally characterize the histopathological changes in the pancreas and kidneys of diabetic rats, which guarantees the use of this methodological tool in other similar models under our experimental conditions.
